Protocol
Immediately upon arrival they were housed in PT cages, and behavior was followed for 14 consecutive days.
Lights were on between 7:00 - 19:00. The first hour of the dark phase (i.e. 19:00 - 20:00) is used as timepoint 1 in all subsequent analyses.
In all graphs, the times on the Y-axis are hours. Hours 1-12, 25-36 and 49-60 are dark phases.


Habituation
During the first 3 days the 12 mice were habituated to the PhenoTyper. After this habituation phase the mice were subdivided into 2 groups (for IP injections and oral administration [PO]), in such a way no initial difference in activity was present between the groups. This dataset is indicated as ?hab?.

Drug administration
Both groups received the drugs at the same time. The first group (PO) received the drug using a ?butter pellet?. The second group (IP) received the drug IP.


1 mg/kg
The first drug dose of 1 mg/kg was administered 15 minutes before the onset of the dark phase (18:45). There was barely an effect of this dose on activity in the subsequent dark phase.

Taken into account that the activity of the FVB mice was already very high during the dark phase in the habituation trial, we came to the conclusion that the effect may not be visible during the dark phase (ceiling effect). In order to make sure the effect is visible for the higher doses, we administered the higher doses during the start of the light phase where activity is in general lower (3 to 4 hours after the onset of the light phase, PCP administration between 10:00 - 10:15, i.e. during hour 40). About 15-30 min after PCP administration video tracking was started. Data from the subsequent entire hour (hour 41) is the first data point in the graphs.


5 mg/kg
At this dose there is a slight effect between the habituation phase and the PCP IP injection of 5 mg/kg during the first hour. There is no effect of the PCP in the PO group at this point. However, when the dark phase starts (from hour 49 onwards), the PO group shows an increase in activity compared to the IP group and the habituation phase.


10 mg/kg
There was a large acute effect on activity during the first hour(s) after IP injection and during the dark phase the animals show lower activity. It may be caused by fatigue, due to the extreme activity during the light phase.
As with the 5 mg/kg dose, the PO mice show a higher activity during the dark phase, although this effect is not as profound.


15 mg/kg
As we noticed the PO group showed no acute effect, and the effect during the dark phase did not change with the dose, the PO group was given an IP injection with saline (group is called Saline (IP) in this dataset) to investigate the effect of IP saline injections in FVB mice.
During previous studies we found there was a large effect on activity when mice (C57BL/6J) were removed from their home-cage and IP injected. Activity typically decreases for several hours (up to 24 hours) in C57BL/6J mice after IP injections.
During this experiment in FVB mice, the IP injection was given during the onset of the light phase, without any effect in the following hours. However, we see a large effect on activity during the dark phase after the injection, which makes it a good control for the previous IP injection groups. Thus, FVB and C57BL/6J mice may respond differently to IP injections.

The acute effect of PCP administration via IP injection becomes even larger with 15 mg/kg. The dark phase that follows shows reduced activity, as we have shown before in the 10 mg/kg group.



Conclusion
With this experiment we have shown that there is a large acute effect (lasting several hours) on activity of when PCP is injected IP during the light phase.

There was no acute effect of PO administration during the light phase. With a lower dose (5 mg/kg) we found an effect of increased activity during the dark phase, whereas there is no effect (or at least not a profound effect) of 10 mg/kg PO.
We also found increased activity during the dark phase for the mice that received a saline IP injection, which was given in the light phase before. This can be an effect of stress induced by removing the mice from the home-cage and the IP injection, in combination with the damage inflicted to their nest when we take the mice of the cage. In reponse to this damage, mice may increase activity (nest building).
Other activity related parameters (time moving, time arrest, time spent outside the shelter, time spent inside the shelter) all show the same effect as the distance moved parameter.
